A Novel Light-Responsive Gene BcCfaS Regulates the Photomorphogenesis and Virulence of Botrytis cinerea

摘要: Light is a fundamental environmental factor for living organisms on earth—not only as a primary energy source but also as an informational signal. In fungi, light can be used as an indicator for both time and space to control important physiological and morphological responses. Botrytis cinerea (B. cinerea) is a devastating phytopathogenic fungus that exploits light cues to optimize virulence and the balance between conidiation and sclerotia development, thereby improving its dispersal and survival in ecosystems. However, the components and mechanisms underlying these processes remain obscure. Here, we identify a novel light-signaling component in B. cinerea, BcCfaS, which encodes a putative cyclopropane fatty-acyl-phospholipid synthase. BcCfaS is strongly induced by light at the transcriptional level and plays a crucial role in regulating photomorphogenesis. Deletion of BcCfaS results in reduced vegetative growth, altered colony morphology, impaired sclerotial development, and enhanced conidiation in a light-dependent manner. Moreover, the mutant exhibits serious defects in stress response and virulence on the host. Based on a lipidomics analysis, a number of previously unknown fungal lipids and many BcCfaS-regulated lipids are identified in B. cinerea, including several novel phospholipids and fatty acids. Importantly, we find that BcCfaS controls conidiation and sclerotial development by positively regulating methyl jasmonate (MeJA) synthesis to activate the transcription of light-signaling components, revealing for the first time the metabolic base of photomorphogenesis in fungi. Thus, we propose that BcCfaS serves as an integration node for light and lipid metabolism, thereby providing a regulatory mechanism by which fungi adapt their development to a changing light environment. These new findings provide an important target for antifungal design to prevent and control fungal disease.

摘要:

The antifungal effectiveness of extracts of five medicinal plant species was determined. The inhibitory activity of extracts of Eucalyptus tereticornisXanthium sibiricumArtemisia argyiTupistra chinensis and Pyrola calliantha were evaluated against the mycelial growth of the plant pathogenic fungi Aspergillus nigerBotrytis cinerea, Penicillium digitatumP. expansum, P. italicumand Rhizopus stolonifer. All plant extracts were prepared at 60°C using solvents (either water, 50% ethanol (v/v), 95% ethanol (v/v), ethyl acetate or petroleum ether). Fungicidal effects of all plants tested were confirmed. Different extracts from the same plant species gave different degrees of inhibition. All aqueous extracts had weak or no activity on all fungi tested. Ethyl acetate and 95% ethanol extracts from T. chinensis rhizomes gave greater inhibition and a broader spectrum inhibition than the other extracts. T. chinensis may have potential as a new natural fungicide and may be used for the preservation of agricultural and forestry products such as fruits and vegetables.

Fusaricidins in

摘要: Four kinds of antifungal compounds from an extract of A21 with molecular masses of 883.56, 897.59, 947.55, and 961.58 Da were characterized as the members of fusaricidin-type of antibiotics according to LC-MS analysis. Fusaricidins isolated from culture filtrate displayed high antagonistic activity against several plant fungal pathogens, especially , the causal agent of gray mold. The fusaricidins biosynthetic gene cluster (BGC) from A21 was cloned by PCR and comparative cluster analysis revealed that gene , the 3′ boundary of the fusaricidin BGC in strain PKB1, was not present in fusaricidin BGC of A21, indicating that is not necessary for fusaricidin synthesis. Fusaricidin extract from A21 significantly reduced gray mold disease incidence and severity on tomato. The mRNA levels for three patho-genesis-related proteins (PRs) revealed that treatment of tomato leaves with fusaricidin extract induced the expression of PR genes to different levels, suggesting that one reason for the reduction of gray mold infection by fusaricidin is induction of PR proteins, which lead to increased resistance to pathogens. This is the first report of the application of fusaricidins to control tomato gray mold and the comparative cluster analysis provides important molecular basis for research on fusaricidin biosynthesis.

Transformation of triclosan by a novel cold-adapted laccase from

摘要: This work investigated the transformation of triclosan (TCS) by laccase produced by a pathogen isolated from rotten tomato. The pathogen was characterized as sp. FQ, belonging to subphylum . The laccase exhibited cold-adaptation with relatively high activity at 20°C. The laccase could effectively transform TCS. Approximately 62% TCS could be removed at dose of 1.0 unit·mL in 120 min. The reaction rate appeared to be to the concentration of the substrate, suggesting the laccase activity remained stable during the reaction. Transformation products of TCS were analyzed by mass spectrometry and it was revealed that TCS dimers were formed via radical coupling pathways. During this process, laccase catalyzed oxidation of TCS to form a radical intermediate is the rate limiting step. However, this step can be reversed by humic acid. Overall, the laccase showed great potential in the treatment of phenolic contaminants. Since laccase is widely presented in natural environment, this study also revealed an important pathway involved in the transformation of phenolic contaminants in the environment.

摘要: This study reports a strain of CCTCC-SBW0162 with potential to enhance biocontrol activity against gray mold pathogen, , and with a pivotal role in tomato ( ) plant growth enhancement. A total of 254 isolates were screened by antagonistic assay. Of these, 10 were selected for greenhouse experiments based on their greater inhibition of . The antagonistic assay and greenhouse experiments indicated that CCTCC-SBW0162 gave the highest inhibition rate (90.6%) and disease reduction (80.7%). Also, to study the possible mechanism associated with antifungal activity of CCTCC-SBW0162 against , molecular docking was used to assess the interactions between CCTCC-SBW0162-derived metabolites, and pathogencity and virulence related proteins of . The molecular docking results indicated that the combination of harzianopyridone, harzianolide and anthraquinone C derived from CCTCC-SBW0162 could synergistically improve antifungal activity against through the inhibition/modification of pathogenicity and virulence related proteins. However, this computerized modeling work emphasized the need for further study in the laboratory to confirm the effect derived metabolites against the proteins of and their interactions.
